چکیده

Breast cancer is one of the most common cancers occurring among women throughout the world, and is the leading cause of death from cancer. Globally, it averages more than 1.1 million new cases, where developed countries face 55% of the burden; the incidence is rapidly catching up in the developing world as well (Parkin et al., 2005). In Pakistan like other Asian countries, breast cancer among females is frequently reported at an advanced stage, which is most likely due to lack of awareness and early screening programs (Ahmad, 1991; Naeem et al., 2008). Being a heterogeneous disease, breast cancer is characterized by multiple subtypes as revealed by differential gene expression profiles. Its diverse nature points towards the involvement of multiple prognostic and predictive tumor markers (Turaga et al., 2010). Epidermal growth receptor 2, commonly known as HER2 is a crucial breast cancer oncogene, which is located on the long arm of chromosome 17 and encodes a 185-KDa cell surface receptor with tyrosine kinase activity (Akiyama et al., 1986; Slamon et al., 1989). Although, early studies have

Imaging features of estrogen-negative breast cancers: a correlation study with human epidermal growth factor type II overexpression

Background: Estrogen-negative breast cancers have different clinical course, prognostic features and treatment response in comparison to estrogen receptor-positive (ER-positive) breast cancers. Human epidermal growth factor receptor 2 (HER2) oncoprotein has found to have a pivotal role in natural cell growth and cell division and is suggested to be directly related to tumor invasiveness in brea...

Prognostic utility of fluorescence in situ hybridization for determining HER2 gene amplification in breast cancer.

An accurate investigation of the HER2 proto-oncogene is extremely important for the therapy and prognostication of breast cancer. Currently, immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH) are standard methods for this purpose. The aim of this study was to detect the expression and amplification of HER2 in paraffin-embedded samples of breast cancer tissue and to investi...

In the era of next generation sequencing, is the cytogenetic analysis still important in polycythemia vera?

South Asian Journal of Cancer ♦ Volume 6 ♦ Issue 2 ♦ April-June 2017 46 showed 19% breast carcinoma with polysomy 17. When we applied 2013 CAP scoring criteria, we found 14.6% increase in number for HER2 amplification with polysomy 17. As we shown one case in Figure 1, polysomy17 with negative according to CAP 2007 guideline but when we applied 2013 CAP guideline on the same case, the case shou...

Analysis of HER2 gene amplification using Differential PCR in breast cancer patients of Isfahan Province

Background: Amplification of HER2 is seen in 20-30% of breast cancer cases. Measurement of HER2 gene amplification appears to be of vital importance in planning the treatment schedule for patients with breast carcinoma. The aim of our study was to evaluate HER2 amplification status in malignant and benign breast tumors by differential PCR (dPCR). Materials and Methods: The genomic DNA was ex...

Predicting Effects of Clinicopathological Variables on Her2 Gene Amplification by Chromogenic in situ Hybridization (CISH) in IHC Her2 (2+) Breast Cancer Patients; A Study from Iran

Background & Objective: The her2 amplification plays an important role in breast cancer management. Therefore, there is a need for using supplementary molecular methods in IHC equivocal cases. Present study has been conducted to determine the effects of clinicopathological variables on her2 gene amplification by chromogenic in situ hybridization (CISH) in IHC ...
